Could someone please explain the best way to remove a siezed spindle.

On the clutch side there is a large nut. Is this left or right thread ?

Its normal right hand thread, there is also an allen pinch bolt on the chain side of the frame and you have to loosen the engine clamps which are on the spindle itself.
Once youve loosened everything the spindle has to be unscrewed from the frame.

The large nut in question is a locknut for the shaft , the shaft screws into the frame on the clutch side . Worst case , remove engine to get a grip on the shaft . Classictrials spindle kit comes with a drilled shaft for greasing and has a allen wrench fitting in end of shaft for installation and removal , and also for adjusting the preload on the whole pivot . Do Not damage any of the pieces on the shaft ! (nut,collar , snapring etc.) as these are just about irreplaceable !!!
